The listing of 4-methylimidazole (4-MeI) on California’s Proposition 65 list has raised interest in the need to analyze 4-MeI accurately and efficiently. Several methods for the analysis of 4-MeI are commonly employed, including LC using UV or mass spectrometric detection and GC-MS after ion-pair extraction and derivatization. This presentation focuses on analysis of 4-MeI using LC with mass spectrometric detection, based on methodology previously developed by Klejdus et al. (J. Separation Sci., 29:378-384, 2006). With the current availability of an inexpensive stable, labeled internal standard, the methodology above can be further optimized. In addition, certain matrices can be analyzed by direct injection after dilution, depending on the LOD desired. Finally, the application of the above methodology to simultaneous analysis of 2-MeI and 4-MeI is also addressed.

Matthew Eckert joined Covance Laboratories in 1999. Over the last 12 years he has held positions as a toxicology dose formulation technician and as a chemist, both in bioanalytical chemistry and nutritional chemistry/food safety.
