Many very subjective methods of analysis have been used to measure flocculation. Using standard yeast growth conditions, the Helm's test was modified to include a comparison of spectrophotometer readings of the culture density after growth vs. a density measurement after the settling reaction used in the Helm's test. The handling of cultures before the test, density of the culture during the settling reaction, and density of the culture during the spectrophotometer readings were determined to be critical parameters for the analysis of routine yeast cultures. Conditions for the direct analysis of brewery yeast slurries are presented.
